Nuclear Factor-KB, an Unappreciated Tumor Suppressor
نویسندگان
چکیده
The notion that nuclear factor-KB (NF-KB) is a tumorpromoting transcription factor has become a widely accepted dogma in biology. However, recent findings suggest an inhibitory role for NF-KB in carcinogenesis and tumorigenesis. Although the tumor suppressor–like effect of NF-KB remains to be rigorously established by further studies using cellular and animal models, these latest findings warrant caution with respect to blockage of NF-KB activation as a broad strategy in treating cancers. Mutant p53 Enhances Nuclear Factor KB Activation by Tumor Necrosis Factor A in Cancer Cells
Mutations in the p53 tumor suppressor are very frequent in human cancer. Often, such mutations lead to the constitutive overproduction of mutant p53 proteins, which may exert a cancer-promoting gain of function. We now report that cancer-associated mutant p53 can augment the induction of nuclear factor KB (NFKB) transcriptional activity in response to the cytokine tumor necrosis factor A (TNFA)...
متن کاملImmunohistochemical Evaluation of Human p53 Tumor Suppressor Protein Content in Ductal Carcinoma in Situ of the Breast
The focus of this study was to determine if early detection of mutant p53 accumulation may be an early indicator of tumor aggressiveness and transformation to invasive breast cancer. For this purpose, the p53 content of 100 human breast biopsies classified as ductal carcinoma (DCIS), was evaluated by immunohistochemical method. All specimens were microscopically classified into histologic types...
متن کاملMolecular and Cellular Pathobiology Nkx2-8 Downregulation Promotes Angiogenesis and Activates NF-kB in Esophageal Cancer
Angiogenesis is a major clinical feature of esophageal squamous cell carcinoma (ESCC), an aggressive disease of increasing incidence in developed countries. In ESCCs, the proangiogenic factor VEGF-C is an independent prognostic factor for ESCC, where understanding the mechanisms of VEGF-C upregulation may cue possible therapeutic insights. Here, we report that expression of the transcription fa...
متن کاملHuman Cancer Biology The Biphasic Role of NF-kB in Progression and Chemoresistance of Ovarian Cancer
Purpose: NF-kB is a transcription factor known to promote tumorigenesis. However, NF-kB is also known to be proapoptotic andmay potentially function as a tumor suppressor, although such a functional role has not been extensively investigated in human cancer. Experimental Design: A dominant-negative mutant of IkBawithmutations at S32A and S36Awas used to inhibit the function of NF-kB in ovarian ...
متن کاملTumor and Stem Cell Biology p53-Dependent Regulation of Mitochondrial Energy Production by the RelA Subunit of NF-kB
Aberrant activity of the nuclear factor kappaB (NF-kB) transcription factor family, which regulates cellular responses to stress and infection, is associated with many human cancers. In this study, we define a function of NF-kB in regulation of cellular respiration that is dependent upon the tumor suppressor p53. Translocation of the NF-kB family member RelA to mitochondria was inhibited by p53...
